Check that the shortcut chosen does not conflict with another. What did you choose?
(isn't it easier to use up/down arrow?)

No there is no conflict. There is no conflict note.
Up and down arrow can not be assigned for some reason.
????
Be aware that this works the same as the toolbar - it toggles the state for the next note you are about to enter, rather than changing the previous note.

This problem occured after I got MuseScore 3.5 BETA Version. May this be the cause?

I mean that in current versions of MuseScore, clicking an acccidental button or pressing the corresponding shortcut does not change the note you just entered. It works like the duration or voice buttons - it changes the next note. Older versions worked different let. So maybe you are simply expecting it to work like it did in the past.
Anyhow, in order for us to understand and assist better, we would need you to describe precise steps to reproduce the problem you are seeing.

I can not write C Sharp
neither On Chord Mode (CTRL + K) nor RNA Mode (SHIFT + R)
I assigned the shortcut at "Edit - Preferences - Note Input : Sharp)
I tried some keyboard shortcut combinations but they don't work.
Version : MuseScore 3.5 BETA

By realizing that entering chord symbols/RNA is not the same as note input.
And then understanding that you don't input sharps into those text, but basic hash/railroad signs such as this #. Likewise, for a flat, you don't enter a flat, but just a lowercase 'b'. For reference though, none of this has changed in 3.5beta compared to the current stable release 3.4.2
