I assume you are talking about one of the 3.5 builds - which one (see Help / About)? And how are you transposing the chords?
There was an issue in earlier builds - maybe included the beta - where what you describe would occur, but I can't reproduce this in the RC, at least not using Tools / Transpose to transpose the full score.
If you are still seeing this using the RC, can you attach a sample score and precise steps to reproduce the problem?

i am using the release candidate and the problem is the same if i transpose by key or interval , the whole piece or only part of it. The first time of the playback (space key) it is still the old key, the second time it is ok

I can confirm using this score. I suspect the key here is, there are no actual notes in the score , that probably is what is preventing the playback code from updating right away. That's just a guess, but I couldn't reproduce once I added a note, and it seems plausible enough. Could you please submit a formal bug report to the issue tracker, with this score as an example, and make mention of my supposition?
