MS4 - serious payback problem

I changed the instrument indicated by the oval and the arrow on the mixer and all the other instruments no longer play in playback. Even clicking on M and S (which are automatically disabled) nothing happens.

Looks like you have one measure selected in the score - that does indeed solo the selected staff. To play all staves, don't select a measure (or multiple measure) - click a single note, rest, or anything else really. Or opress Esc after selecting a measure.
