I think you probably are looking at the buttons that you would use to select which voice you are inputting notes to. See https://musescore.org/en/handbook/3/voices for what that is all about.
So, in generl, unless you are deliberately trying to use multiple voices, the 1 should always be enabled while in note input mode - that's the normal default voice. If you are having problems with MIDI input, it probably is not related to those controls
