It isn't clear what you mean by "transpose" here - are you saying you alreayd entered the notes in the wrong key, and now you want to correct that error? If so, then yes, you need to use Notes / Transpose. But I'm thinking maybe you haven't actually entered the notes yet, meaning you don't need to transpose anything at all you simply need to add the key change. To do this, simply add if from the Key Signatures palette. Add it to the first measure you want it to apply to - the first line of the next system. The courtesy signature shown in your picture will appear automatically.
If that doesn't answer your question, please attach your actual score and explain in further detail what you are trying to do.
