Playback
Previously I was able to play .mscz files using the playback buttons.
Now it will no longer do this, either on old files that previously worked, or new files. The buttons are greyed out
I note also the View -> Synthesiser option is also greyed out. I attempted to reset to Factory Settings as suggested in some forum page, but this made no differene.

None of the pull downs contain any options. Actually appear to be disabled.

I downloaded MuseScore 2.2.1 which I already had and then installed in 'repair' mode.
The problem is not solved.
View -> Synthesiser is greyed out, so I cannot navigate to "Fluid"

Open Synthesizer and add the default soundfont again and/or revert to factory settings?

If Synthesizer is greyed out, reverting to factory is the only remaining option.
Hmm, maybe on that preferences page you can revert its settings to default.
On Windows is should be API: MME and Device: Microsoft Soundmapper - Output
Changing the settings there need a restart of MuseScore to take effect

Synthesizer is grayed out if you don't have a score open as well, so make sure you've done that first.
I also attempted to export as MP3 which previously worked in other cases, but attempting to play back I got the message: " This item was encoded in a format that's not supported"
Investigating further, it appears to be a problem with some Windows 10 upgrade, as I tried to play a previously working MP3 file which caused the same problem.

What version number of Windows 10?
I'm running 1709 (aka Fall Creators Update).
Also, how did you try to play the previously working MP3?
Certainly not on MuseScore...

Yes you are correct certainly not with MuseScore. What I was trying to say was that it no longer appeared to be a problem with MuseScore becauses I was attempting to play an MP3 file using another App.
Anyway problem solved, it was a bad upgrade to Windows 10 which has since been corrected.
I now have Windows 10 Home version 1803. OS Build 17134.48

Reports a broken "Microsoft print to PDF" which formerly worked before the update.
(I'm still running version 1709 -- for me, the Settings update status says: 'Your device is up to date'.
So, hopefully, by the time 1803 rolls out to me, I won't have any problems.)

In case of interference with other programs, esp. on Windows, go into your soundcard configuration from Control Panel and ensure that the "Allow applications to take exclusive control of this device" in the Advanced tab of both Playback and Recording is unticked.

Many thanks for your further interest in this.
I contacted Microsoft "Feedback Hub" to discover that a recent 'enhancement' to Windows 10 had caused this problem. I have since received an upgrade from Microsoft and all is working well again. It was never a fault with MuseScore (my apologies for suggesting it).
