I have many scores with first and second endings and repeats, but MuseScore now plays through them without regard to repeat signs.

Plays and repeats just fine here.
Make sure the "Play Repeats" button in the toolbar (looks like an end repeat barline) is pressed. If it is is, toggle it, off and back on.
Repeats in that score work fine for me. Most likely you accidentally disabled playback of repeats. See https://musescore.org/en/handbook/3/play-mode#playback-toolbar for how to re-enable it.
