oops my mistake, I didn't think the upload had worked. Do you know the answer?

Which sound font are you using? If you didn't change the mixer it's the first one listed in the synthesizer under the fluid tab.

I tried to use timbres of heaven but i'm not even sure it worked. Even when I try to use the default soundfont and then "save to score" nothing happens

There have been such reports before, see
https://musescore.org/en/node/308905

and there's a fix that went in 3.5.1 that might be related, to do with Single Note Dynamics and some code refactoring that caused issues. I may be entirely wrong on this though
Maybe #309727: Accent playback inconsistent - too harsh for piano, too subtle for flute

Save to score only saves the current sythesizer settings to the score so you can later click load from score to return the synthesizer to that state. What matters is what is in the synthesizer when you press play. The synthesizer can only have one setup at a time and it applies to every score. If the problem is with Timbres of heaven then you would need to report the problem to http://midkar.com/soundfonts/ where the sound font is developed. This is if the info Jojo posted doesn't help. Version 3.5.1 will be released in the next couple of days if you want to wait to see if it's fixed there. With the release of 3.5.1 being so close I'm not going to try to get you to test a build that's not meant for release because the folders are different than for a release build.
Which note sounds sharp to you? The D's in the clarinet in bar 8? I don't hear anything particular off. A tuner shows them as a few cents off of strict equal temperament, but wind instruments usually aren't in strict equal temperament to begin with, so this sort of variance is pretty normal. In particular, that D would be expected to be 1.5X the frequency of the G a twelfth below, and there's no way then that both notes will adhere to strict equal temperament. All wind instruments present their own compromises, but clarinet perhaps uniquely so because of the way that register key works.
